Carlo Ancelotti's side will be out for revenge following their last meeting with Barcelona, a 3-1 defeat in the Supercopa final in Saudi Arabia back in January.
For the first time in the league this season, neither side managed to win their games on the weekend, leaving both sides ruing missed opportunities to either close the gap or extend their lead on the other in the table.
Real Madrid needed their 18-year-old striker Alvaro Rodriguez to score his first goal for the club in just his third game in the 85th minute of their Madrid derby against Atletico to snatch a point against Diego Simeone's men.
That draw gave Barcelona the chance to go 10 points clear at the top of La Liga with a win over 17th placed Almeria, but fell to just their second league defeat of the season in what Xavi and his side will see as a huge opportunity missed.
The defeat in Almeria leaves Barcelona in the unfamiliar position of having now lost their last two matches, having been knocked out of the Europa League by Manchester United last week.
Barcelona have won five of the last eight Copa del Reys, and will be gunning for their second domestic title should they get the better of Real Madrid over two legs, where the winner will play Athletic Bilbao or Osasuna in the final in Seville.
Despite their dominance across Europe and Spain in recent years, the Copa del Rey has evaded Los Blancos in that time, with their last appearance in the final coming back in 2014 - the famous final which was won in the dying moments by that Gareth Bale goal.
